ABSTRACT

Objective To explore the changes of the expression of islet amylodi polypeptide ( IAPP ) and somatostatin( SS) of islet in type 1 diabetes mice, and the mechanism of the expression changes .Methods We established the diabetes model in C57BL/6J mice by low-dose streptozotocin ( STZ) injection.The excisions of the pancreas tails removed on the 3th, 7th, 10th, 14th, 21th and 28th day.Tissues were assessed by immunohistochemical SABC, immunofluorescence in the study .Results 1.Numerical density on area ( NA ) of IAPP positive cells in experimental group (EG) decreased since the 3th day, but average absorbance increased since the 7th day.2.NA of SS-IR cells in EG increased since the 3th day, and average absorbance increased since the 7th day.3.The results of immunofluorescence double staining showed that , IAPP and SS could coexpression in part of cells in islets .Conclusion The number of IAPP positive cells in type 1 diabetes is decreased , but the immunoreaction increased .Immunoreaction and number of SS positive cells increase .Both of them are involved in the pathogenesis of type 1 diabetes.

ABSTRACT

Objective To investigate the effect of the heroin addicts on rat's thyroid C cells, the secretion of calcitonin(CT) and serum FT_3, FT_4, TSH. Methods Adulthood 55 male SD rats were randomly divided into normal control group(NCG), saline control group(SCG) and heroin dependence group(HDG). Heroin-dependent rat models were set up by the way of subcutaneous injection of heroin and the thyroid glands were excised on the 10th, 17th, 24th, 31st, and 38th days after models were set up. Tissues were assessed by immunohistochemical SABC method, cell counting, image analysis method and chemiluminescence techniques. ResultCompared with the normal control group(NCG)and saliane control group( SCG), the immune staining intensity was weakened;the number of CT positive cells was reduced(P<0.05);the average gray value was significantly higher(P<0.05) and showed a gradual increase in the CT positive cells of heroin dependence group(HGD). The concentration of TSH in HDG was not statistically significant as compared with the NCG. The concentration of FT_3 in HDG decreased gradually on the 10th day and the 17th day as compared with the NCG.(P<0.05). The concentration of FT_4 in HDG increased gradually on the 24th day and the 38th day as compared with the NCG.(P<0.05). Conclusion During heroin dependence period, the number and functional of CT positive cells has decreased. The changes of concentration of FT_3 and FT_4 suggest that the function of thyroid gland has changed.

Objective To observe the effect of heroin on EGF and IL-2 positive cells in the submandibular gland of rats. Methods Normal SD rats were divided into normal control group (NGC),saline control group (SCG)and heroin dependence group (HDG). The heroin dependent model of rats was established by subcutaneous injection of heroin, and submandibular glands were excised on the following days respectively: the 10th, 17th, 24th, 31th and 38th. ABSTRACT

Objective To investigate the ontogeny of the islet amyloid polypeptide immunoreactive (IAPP IR)cells and 5 hydroxytryptamine(5 HT)immunoreactive cells(EC cells)in human fetal small intestine and the relation between IAPP and 5 HT. Methods The immunohistochemical technique was used. Results IAPP IR cells were only observed in the fetal duodenum.At 16 weeks,single IAPP IR cell occurred in the epithelium of duodenum;From 22 to 27 weeks,the number of IAPP IR cells was gradually increased,which were mainly located in the intestinal glands.EC cells were detected in each section of small intestine,with the growth of fetus,the density of EC cells in small intestine decreased in order of duodenum,jejunum and ileum respectively.At the week of 11,EC cells were found in the epithelium of intestinal villi and undifferentiated glands.The number of EC cells showed a peak from 17 to 21 weeks of gestation.EC cells were dominantly concentrated in the epithelium of root villi and glands.After 22 weeks,EC cells decreased.The co existence of IAPP and 5 HT was not detected in the same cell,as compared with adjacent sections.Conclusion\ The results indicated that IAPP and 5 HT were expressed in endocrine cells of human fetal small intestine.IAPP IR cells and EC cells varied with the development of the human fetus. ABSTRACT

Objective To investigate the ontogeny of the islet amyloid polypeptide(IAPP)and explore the relationship IAPP and other classical hormones in human fetal colon and rectum. Methods The localization of IAPP immunoreactive(IR) cells and 5 hydroxytryptamine(5 HT) IR cells in 31 human fetal colon and rectum from 9\|27 weeks of gestation was investigated with immunohistochemical SABC method. Results At 9 weeks of fetal age,a lot of 5 HT IR cells were found in the colon,but IAPP IR cells appeared at 18 weeks.In the rectum,both 5 HT and IAPP IR cells were present at 11 weeks,the number of 5 HT IR cells ascended gradually with fetus aged,reached a peak at 20 weeks and dropped gradually after 21 weeks,while in the whole fetus period,IAPP IR cells appeared fewer and scattered.The co localization of IAPP and 5 HT in some cells of colon and rectum was proved as compared with adjacent sections.Immunohistochemical double staining procedures on the same tissue section also showed that IAPP co existed with 5 HT in some cells.Conclusion\ During the human fetus,IAPP were synthesized and co existed with 5 HT in endocrine cells of the colon and rectum.The possible function on the co existence of IAPP and 5 HT in the colon and rectum of fetal was discussed. ABSTRACT

Thyroid glands from 80 human fetuses, 7~31 weeks in gestational age were obtained to study the morphological development of thyroid glands by histochemical, immunohistochemical and TEM techniques. During 7~8.5 weeks the glands composed mostly of epithelial cell cords and abundant glycogen were seen in the cytoplasm of the cells. Some follicles and small accumulations of colloid in the center of the follicles appeared at 12~12.5 weeks and less glycogen was found in the cells. C cells were distributed mainly in the posterior region of the upper pole of both lobes, they usaually occurred singly or in small groups. During intrauterine life, C cells occupied three positions. They are found between follicular cells, between follicular cell and basement membrane and between follicles. But they are situated predominantly in the follicular wall. Three morphological variants were found in the human fetal C cells: (1) spherical or oval cells, (2) polygonal or pyramidal cells (3) cells with a cytoplasmic process. Developing C cells seem to have few secretory granules by TEM and argyrophilic reaction in semithin sections.

ABSTRACT

In this article 108 human fetuses from 12 to 30 weeks of gestation, 3 neonates and a 1.5 years child were used for study of the histogenesis of the appendix.In 12 weeks old fetus, the appendix showed small lumen and thin wall. The mucosa presented intestinal villi and crypts. The endocrine and goblet cells could be found. The circular muscle and serous membrane had differentiated. In the specimens of 13 to 15 weeks, the lamina propria contained diffused lymphatic tissue and lymphatic follicles. On such sites, the intestinal villi were absent. The surface epithelium became attenuate and composed of cuboid or lower cuboid cells. The outer longitudinal muscle layer was evident. The scattered muscularis mucosa was found. At 16 weeks, the endocrine cells appeared and showed various forms. The open-type or closed-type endocrine cells could be distinguished easily. Under EM, based on the morphology of the basal granules, the endocrine cells might be divided into two types. The granules were cell enclosed in a limiting membrane. One type of the granules was round and various in size, and another type was irregular. In the surface epithelium covered on the lymphatic tissue, the microfold cells could be found. They showed irregular microvilli or mierofolds and numerous apical micropinocytotic vesicles. At 18.5 weeks, the villi tended to merge each other. At 20 weeks, the muscularis mucosa was formed and the 4 layers of appendix resembled those of the normal adult. We found endocrine cells aggregated into Segi's cap on the apex of the villi in 7 fetuses of 16-21 weeks. The T or B lymphocytes could be identified as early as 13.5 weeks by immunologic method. We found the percentage of the B lymphocytes decreased with the gestation age, and this fact suggested that the appendix might be a B lymphocyte pool during the fetal period.

ABSTRACT

Objective To observe the expression and localization of the islet amyloid polypeptide(IAPP) in the stomach of human fetus. Methods The localization of IAPP immunoreactive(IR) cells in 12 human fetal bodies of stomach from 11th to 20th week of gestation was investigated with immunohistochemical SABC method. Results At 11th week of human fetal age,the IAPP-IR cells had been found in the gastric mucosa.The IAPP-IR cells were dominantly concentrated in the fundic gland.The number of IAPP-IR cells increased gradually,and the immunostain of immunoreactive cells darkened gradually from 14th to 20th week of gestation.As compared with the adjacent sections which were stained by HE,some IAPP-IR cells were co-localization with parietal cells.Conclusion IAPP-IR cells exist in the human fetal body of stomach..
